PCdb: A comprehensive plant genome editing database integrating sgRNA efficiency, off-target predictions, and epigenomic landscapes
The off-target effects of gene editing encompass both DNA sequence alterations and chromatin modification changes. DNA repair mechanisms at off-target sites may introduce epigenetic modifications. The rapid advancement of genome editing technologies has led to the development of numerous tools for identifying and scoring off-targets across entire genomes. We present PCdb, an interactive and bioinformatically enhanced platform for analyzing off-target cleavage, aimed at enriching gene editing safety analysis. PCdb contains an extensive collection of functionally validated single guide RNAs (sgRNAs), enabling precise identification of off-targets. Furthermore, it incorporates epigenetic markers at off-target sites. As the first curated database of its kind, PCdb promises to enhance safety quantification research, inform experimental design, and drive the development of computational off-target prediction tools. It features a user-friendly interface that facilitates data browsing, querying, analysis, statistical evaluation, and data export.
